There should the return air duct be place in a room 25' x 20' , 9' high for cooling and heating?

If the supply is place over head will one air return work (per supply) and can the return air duck be place in the ceiling.

One return should be sufficient for this size room.  It should be located as far away as possible from the supply to prevent "short-cycling".
