  1. depends on where you start from, the Canal Grande runs in to the Canale di San Marco to get to Murano.  You want the water bus # 62 to Lido and Murano


  2. Venice and the other islands like Murano are located in a Lagoon.

    So, when you leave Venice to go to Murano, you won't cross any particular canal - even if, more precisely, there will be the Canale delle Fondamente Nuove between that side of Venice and the San Michele island, where the Cemetery is located.
