What are the requirements for a species to be analyzed using spectrophotometric method?

  1. It must show an absorbance in range of wavelength the photometer uses.

    That is it has to be colored, if you use the visible range. And it has to absorb in the UV-region if you use a photometer below let's say 400 nm.

    And, I nearly forgot, if you use a transmission type photometer the sample analyzed has to be transparent for the applied wavelength.
