it really depends on where you're buying it and how old it is.  new car?  take it out and see if you like the way it drives.  used car (less than 10 years)  pop the hood and make sure it all looks in order.  check for oil on the engine and rust on parts.  aside from the obvious, the best way to find problems with a used car is to go out in the country and act like you're at a drag race.  floor it to about 80, observe any sounds or vibrations that seem out of place, then stop the car as fast as possible.  that will usually tell you if something major is wrong with it.
