okay. firstly, ignore a lot of the c**p that people are goin to say about cancer etc. yes, it *could* be cancer. but that's highly unlikely.

Most likely is epididymitis - infection of the epididymis, which is what connects the 'tubes' to the t******e. This can be caused by quite a few bacteria and the such, and he'll need to go to a doctor to get a urine test.

It could also be orchitis, where the epididymitis has spread to the t******e, which causes further pain.

If he ever gets any discharges (blood, funny colored material or whatever), then he needs to see a doctor because there's definately an issue.

It could also be testicular torsion - which is treated as a medical emergency. This is when the 'tubes' get tangled and twisted, which cuts off blood supply and may lead to infertility.

It could be many other things - varicocoele, hydrocele, cysts in the epididymis or hydatid of morgagni, spermatocele.

He should see a doctor, and they will probably give him a referral to have an ultrasound (to check out any developmental issues or screen out testicular torsion or potential cancer) plus a urine test to check for orchitis/epididymitis/urethritis (essentially a UTI).

I could lead to more serious matters.  What we don't know is if he's got water on the testicles or if there's been some kind of muscular or tendonous damage to the area.  Any of these will cause a great deal of discomfort.  The fluid build-up part being the worst.  A friend of mine from the military had a fluid build-up on his testicles that he thought was just his need to *ahem* take care of business.  He ended up having to have the fluid drained directly by the doctor.  I can't say how it happened...it just did.  He didn't remember injuring himself in any way.  

Whichever the issue, he should consult the family doctor to get it taken care of.  It's not likely to be one of the issues that takes care of itself after having intercourse, per se.
