    Nuclear stains are carried out on bllod smears to find a variety of antigens most commonly auto-antibodies and proteins.

    Depending on the method used...immunohistochemistry, immunofluorescence or silver staibs (AgNOR)....the pattern of nuclear staining is noted and scored.

    The scoring depends on 2 factors....

    1. strength (negative 0,  weak positive also called1+, moderate staining 2+ and strong positive 3+). The strengt is usuallycompared with a known standard (positive control).

    2. Pattern ofstaining - diffuse (throughout the nucleus uniformly) or speckled (grainy staining - like black dots in a white background, some therefore also call it - salt and pepper appearance)

    Since you did not mention what test was performed on your blood..i cannot comment further,

  3. ANA (antinuclear antibody) testing of the serum is read by microsopic examination of the staining of cells onto which the serum has been applied.

    If the serum has antibodies that attach to the cells, then staining the cells with the right stain will make the part flouresce under ultraviolet light that has the antibody attached to it.

    "ANA shows up on indirect immunofluorescence as fluorescent patterns in cells that are fixed to a slide that is evaluated under a microscope. Different patterns are associated with a variety of autoimmune disorders. Some of the more common patterns include:

    Homogenous (diffuse) - associated with SLE and mixed connective tissue disease

    Speckled - associated with SLE, Sjogren’s syndrome, scleroderma, polymyositis, rheumatoid arthritis, and mixed connective tissue disease

    Nucleolar - associated with scleroderma and polymyositis

    Outline pattern (peripheral) -associated with SLE " (below)
