It means the volume of air and fuel an engine can take in during one complete cycle of the engine.  To put it another way, if all of the cylinders were cups, it is how much volume they can hold in all of the cylinders from bottom dead center to top dead center (the highest and lowest points the pistons move).

A liter bike is A sports motorcycle with an engine displacement of about 1,000 cc or more.  A liter is approx 61.02 cubic inches.  If you convert that to metric, it will be approx. 1000 cubic centimeters.  Therefore, you have a "Liter Bike."  They are the larger of most of the supersport motorcycles.
