I don't have personal experience at 24 Hour Fitness, but I have interviewed for a front desk position before. Common questions are going to be about your experience working with the public, your problem solving skills, and depending on how this place is set up, your comfort level answering phones, doing paperwork, and working computers.

To prepare for an interview, I recommend thinking over your skills and experiences. It always helps me. If you have some sort of idea of what you want to say already in your head, then it can make answering the questions a lot easier. During this interview, you also need to make sure you demonstrate your ability to interact well with people you don't know, and show how comfortable you are working with the general public.
