  1. The cyanohaemoglobin method -

    Using a pipette add a sample of 20 µl of blood to 5ml of Drabkin’s solution in a test tube and mix thoroughly.  

    DRABKIN’S SOLUTION:-

    200 mg Potassium ferricyanide

    50 mg  Potassium cyanide

    140 mg  Potassium dihydrogen phosphate      

    Distilled water made up to 1 Litre



    Place approximately 2ml of the resulting solution into a cuvette, and read the absorbance values in a spectrophotometer at 540nm.

    The haemoglobin concentration of the blood sample can be calculated from a curve prepared from known standards.
