  2. The answer above is incorrect only in one detail - protein kinase C does not phosphorylate tyrosine residues - it's a protein serine/threonine kinase.

    There is a family of related protein kinase C genes, but the enzyme referred to as protein kinase C was originally discovered by virtue of its ability to be activated by phorbol ester tumor promoters.  In a cell, this common and abundant protein kinase is activated by diacyglycerol and membrane phospholipids, in the presence of elevated calcium.  Some subtypes of protein kinase C do not require the elevated calcium, and are simply lipid-activated.
