Small block Chevy all the way.  I like a stroker, 383, on a 4 bolt block.  You can take an SBC all the way up to 427ci and it has a much better power to weight ratio than the big block.  That's why so many hot rodders have used them over the years.

As for transmission, I put a Tremec T-56 in my most recent build.  It's a 6 speed with two overdrives, so you get basically a close ratio 4 speed with 2 loafing gears for the highway, so the noise won't drive you crazy.  I bought one out of wrecked TransAm.  It takes a couple of tricks to run it without a computer controlled setup, but nothing too hard to overcome.
