Kind of depends on what you mean by 'role-playing board game', since those are normally two different types of game.  If you want actual roleplaying games, D&D is by far the most popular, though I would wait for the upcoming 4th Edition (coming out in June) which looks to be a streamlined and easy to play version.

There are a ton of other roleplaying games out there; but very few of them offer good intro sets that are easy to learn.  I am fond of Big Eyes, Small Mouth (2nd Edition Revised, sadly now out of print) and Savage Worlds (Only $10 for the core rules!  Or you can try out a sample version with a free download - check greathwhitegames.com for it) for versatile games.  Both much easier and more flexible than GURPS.

If you actually want a boardgame with classic RPG elements, you might check out Fantasty Flight games line of games such as Descent, a dungeon crawl adventure or Arkham Horror, a cooperative game pitting the players against horrors from beyond space and time based on the works of HP Lovecraft.  Another good, and very easy to play boardgame that feels a bit like an RPG is Last Night on Earth: The Zombie Game, which feels much like an action-filled zombie movie to play.

Best is a matter of subjectivity. The most widely played is Dungeons & Dragons, of course, the game that made the hobby popular. The most versitile is Steve Jackson Games' GURPS. The game has basic rules and you can buy books that adapt those rules to hundreds of genres from fantasy to scifi to super heroes.
