I agree with sarah about Seoul.  It has everything, including history.  There are a number of great museums and historical sites (700 year old palaces and gates) that are very cheap to visit.  Transportation is excellent and not too expensive.  There's great shopping and plenty available in terms of dining and hotels.

If you want to spend time outside of the big city, I've been to Jeju Island, Pusan, Pohang, Taean, Daejeon, Gwangju, etc., and the best place by far was Gawangdo Province: Gangleung Beach and Sokcho (town near Sorak Mountain).  The air is clean, people are REALLY friendly, the food was much better than Jeju, and the water was just as clear.  Jeju is also more expensive and touristy.  We went to jeju and were were just tourists, but in gawando, people wanted to hang out with us and show us where the best restaurants are.  It was the BEST weekend I had in Korea.

Because of its beautiful and pure sea, many people in Asia visit the island.

Seoul will probably the best tourist destination.

Not only can you experience the historical traditonal aspects of the South Korean culture, but you could also experience the modern culture as well. There's thing street fashion mall that's popular for teenagers, young adults. It's in seoul and the town's name is : Myung-dong

If you probably nature/scenery or historical asian archietect places, Pusan will be good also. However you could find this Seoul as well.
