The initial irony is that Juliet is praying for the coming of darkness of night so that Romeo will come.  She is unaware that Romeo has slain her kinsman Tybault and that their plans are certain to go awry.  She suggests that when Romeo dies night will make him into many stars and make the heavens more brilliant than the day.  This is foreboding his death though she thinks she is being romantic.  Darkness and night are usually dreaded and the news coming is to be dreaded though she doesn't know that yet.

The next major irony is that Juliet thinks the Nurse is talking about Romeo's death when she comes in and starts telling Juliet about Tybault's death.
