A genomic library potentially contains the entire genome of an orgaism fragmented into small pieces and inserted into some type of cloning vector such as a bacteriophage or a plasmid.

Once you have the library constructed, you can begin to search for genes (or other regions of the genome) of interest.  Once you have identified the phage/plasmid carrying the sequence of interest, you can then grow large amounts of the phage/plasmid containing your DNA for study, cloning, etc.
