If you are talking gas powered largest displacement, from the factory it was the 1970.s Cadillac line with 500.1 cubic inch displacement (CID).  There are also Hemi powered drag cars and boats up to 572 CID.  In the diesel world, Semi trucks can go to 15 liters (around 900  CID) and trains are powered by larger versions of that design.
