there is no online booking for rameswaram to kanyakumari ,

you have lot of bus there and you need not worry for that .

if you want good road and fast service then do not go via short route as the road is not good .

you can come to madurai from rameshwaram . every 15 minute you have a bus to madurai and from madurai you can come to nagerkoil /kanyakumari here also you have every 10 minute a bus. the government bus are very good in this both route and also safe and comfort , as there is no private bus in the tamilnadu excepet omni bus which runs mostly in the night my preference will be this only. the total journey hours is 10 hours from rameshwarm to kanyakumari.you have night service also from rameshwaram to kanyakumari via madurai.
