It takes place in the fictitious town of St. Petersburg, Missouri, patterned on Twain's home town, Hannibal, some time before the Civil War.  Since Twain was born in 1835 and the story is based to a large extent on his childhood, it seems reasonable to suppose that it takes place in the 1840s..
