  1. Ok I just did this last semester- the purpose of the LB plate is to see the bacteria's growth without antibiotics and the LB-amp plate is used to see how the bacteria can grow with ampicillin with the pGLO.  So it will allow you to see how the pGLO allows the bacteria to grow with ampicillin and how well it grows with the amp and pGLO compared to the one without amp.
