  1. Isolated chlorophyll in a tube fluoresces when light is shone on to it  - the reason is that the light provides energy to electrons in the chlorophyll which as a result are raised to  higher energy level in the chlorophyll molecule and when these electrons are fall back to their original level the absorbed light energy is released as red  fluorescence.

    However when the chlorophyll in intact chloroplasts absorbs light they pass their electrons on to electron carriers in the thylakoid membranes (light stage of photosynthesis) resulting in ATP & NADPH production rather than red fluorescence.
