When is it safe to shift job after i485 filled and 140 not approved?

Your Immigrant Worker Petition has been denied.  You do not qualify to adjust status or get a green card (permanent residence) based on your employment.  As soon as your H1-b or whatever employment visa you're on expires, you must leave and return home.  

You cannot change jobs unless you return home, then get hired by a firm willing and able to obtain an employment visa for you.  Since your 140 has been denied, it is questionable whether you can get approved for another work permit.
