I check with the post office and then I figure what does it cost to drive there, and some also charge for the materials. After you ship a few items you will get the jest of what it cost for certain package sizes.

eBay has a Shipping Wizard that will help you calculate costs as you post your listing.

The easiest way to figure it out is to search for the item you want to sell, and look at what others charge for shipping. That's how I started. After I sold a few things, I saw that others seemed to charge a bit more than I did, and so I lowered my shipping (making sure my costs were covered), and was able to get more bids because of my lower shipping.
