  1. Renin is produced by the kidneys in the juxtaglomerular (JG) cells as a result of reduction in tubular NaCl. It is released into the bloodstream where it acts upon a circulating substrate, angiotensinogen (released by the liver) that undergoes proteolytic cleavage to form the decapeptide angiotensin I. Vascular endothelium, particularly in the lungs, has an enzyme, angiotensin converting enzyme (ACE), that cleaves off two amino acids to form the octapeptide, angiotensin II (AII) which is a potent vasoconstrictor raising the blood pressure.

  2. Renin-Angiotensin system plays a major role in blood pressure management ,balance of cardiac volume and salt balance as well as in physiological and pathological process due to stimulation of local Renin receptors.

Receptor is abundantly expressed in heart, brain, placenta and eye and comparatively lower expressed in liver and kidney.

In normal human kidney and heart ,it is localized in mesangium and in the coronary and renal artery ,associated with smooth muscle cells.
