The Smithsonian, although it's not just one museum, it's several.  I think the Louvre is probably the largest one building museum.

Most people think its the smithsonian but that is more than one museum. I think the largest is the louvre in Paris.

The Smithsonian is the largest entity, but has dozens of museums.  The Hermitage is the largest, with five huge buildings in St. Petersburg.  I would think the Louvre might be second.
