If I had to pick just one, I would say quantum mechanics.  It has revolutionized our understanding of physics, and it is what is allowing us (and will continue to allow us) to develop much of the exciting new technology that we have today.

I would say DNA. The human genome project was the basis of most all the development in biological studies. We have better forensics, oran transplantation, making of plastic organ, genetic engineering etc etc etc due to the better Understanding of DNA.
