If you're talking about matching a person's DNA to another sample, as forensic scientists use to place a suspect at a crime scene (so-called "DNA fingerprinting"), the technique was devised by Dr. Alec Jeffreys (now Sir Alec Jeffreys) of University of Leicester in the UK, in 1984. Like many great scientific discoveries there was a certain amount of serendipity to it - he was experimenting with DNA tests on a lab technician and his family, searching for the gene responsible for human myoglobin production (a protein found in muscle tissue), when he noticed similarities in the patterns produced on the X-ray film and had what he called a "eureka" moment - these patterns could be matched up to prove a person's identity and genetic similarity to related people.
