  1. Ultra low temperatures are needed are needed for a variety of laboratory techniques such as Freeze-fracture (a method of tissue preparation for microscopy).

    When saving biological material liquid nitrogen is often used to prevent degradation of tissue (instantly stops enzymes). For example if you want to harvest leaves (or any other tissue) for later mRNA isolation you must freeze in liquir nitrogen if you want a good sample. Likewise flash freezing in liquid nitrogen is necessary when freezing cell cultures you wish to grow later (yeast and bacteria). The flash freezing leaves the cells intact where slow freezing will rupture more cells.


  2. because it is a cryogenic fluid (compressed nitrogen)! it helps in preservation of the living tissue . liquid nitrogen takes heat from d tissue to cool it n hence it gets a very low temperature as low as -219 degree centigrate in case of rbc preservation.

  3. Probably to freeze and store tissue samples for later testing? Or cryogenics use?
