Both are permanent observers, so while they do not vote they do get to participate/observe in certain capacities.  

The Vatican (Holy See) is not a full member because it chooses not to be.  Originally, this was because the Holy See doesn't actually operate as a country.  It's military/security is provided by the Swiss and it's located inside Italy.  It's the ruling organization for a religious organization and while it operates as a nation-state in many ways, it is unique.  Recently, the Holy See has chosen not to be a U.N. member, but it has the full rights of a member of the U.N., except for voting rights.  

Taiwan is not a full member because of China.   In order to be a member of the U.N., you have to be an independent nation state.  The People's Republic of China considers Taiwan to be a part of the PRC.  The position of Taiwan once agreed with this and both the gov't of Taiwan and the gov't in Beijing claimed to be the rightful government of China.  (Taiwan, the U.S., and other allies now consider Taiwan to be an independent state.).  In the aftermath of the revolution, when it became clear that the rulers in Beijing controlled the majority of China and were functioning as that nation's government, the U.N. took the Chinese membership from the government in Taiwan and gave it to the government in Beijing.  Now as a permanent member of the U.N. Security Council, and as a major economic power, the government in Beijing has effectively blocked any attempts to award Taiwan membership in the U.N.
