  1. Gram stain is designed to chemically react with the cell wall of active bacteria, thus producing the colors seen under a microscope.

    There are many different kinds of spores produced by everything from protozoan parasites to trees, but they all have one thing in common. They are enclosed in a relatively impervious coating case of some sort, which protects them until conditions are right for whatever is the next phase of their life cycle. These coatings are not cell walls and would therefore likely not react with gram stain. Depending upon the type of spore you are looking for, there may be a specific 'spore stain' or not. Such a stain would be designed to react chemically with a particular type of spore case, which would then produce a visible color change under the microscope.
