For File Services / Shares, it's not that complicated. Just :

1. right click on the folder you want to share,

2. hit Sharing and Security,

3. choose Share this Folder,

4. Click on Permission.

5. Then from there you can now add Group or User from your Domain.

6. Then choose the appropriate permission for each group or user.

It's easier if you organize your domain users by creating groups, this way you can give permissions by group especially if you have a lot of domain users.

Another powerful tool that you might want to explore is the Group Policy. I have been using this for a long time starting with windows 2000 Server, and it really saves me a lot time. It takes time to setup the policy but it pays off in the long run.
